Assignments:

-Answer the pGLO questions on the handout that came with the lab

- Read Biology book 13.2. to learn about restriction enzymes (do mini-lab on p. 343, problem solving lab p. 347, Section assessment p. 348),

- solve plasmid cutting worksheet,

- make your own worksheet from NEB website (Go to NEB website and use the cutter tab to create a new worksheet for me. Choose a plasmid, cut it with at least 3 restriction enzymes (they should make at least 4 total cuts – more is great too), show the sizes of fragments when cut with each enzyme and each combination of enzymes. Make me a worksheet like the one I gave you and make an answer key for the cuts. Be sure to identify the plasmid and the enzymes.)]–    

- find the 1 cutter enzymes that will cut the GFP gene from the pGLO plasmid. Predict the fragment size.

-Restriction enzyme lab

            *Grow the bacteria with the pGLO up and use the mini-prep kit to remove the plasmid.

            *look at the enzymes we have and predict the fragment sizes when cut with 3 enzymes in every                                   possible combination. Draw out lab directions. Cut the plasmid with the 3 enzymes and all                  combinations of them.

            *make and run a gel with a ladder to see the fragment sizes.

-design the primers that would be needed to use PCR to make many copies of the GFP in a pGlo plasmid

-test over the information when the labs are done
